The studies in the Tardiff laboratory utilize an integrative physiologic and biophysical approach to delineate the underlying links between structural and functional alterations in cardiac thin filament proteins and the resultant complex cardiovascular phenotypes observed in patients with Hypertrophic Cardiomyopathy (HCM), a disorder affecting 1/500 individuals. Hypertrophic Cardiomyopathy is an autosomal dominant genetic disorder and is one of the most common causes of sudden cardiac death in young people. Work from our lab and others has shown that the molecular effects of independent gene mutations in sarcomeric proteins are varied and disrupt both sarcomeric function and myocellular signaling pathways leading to the complex, often progressive cardiac remodeling that characterizes HCM. While the first gene mutations linked to HCM were published over 20 years ago, many of the basic mechanisms underlying the disorder remain unknown. It is our goal to elucidate these mechanisms by combining techniques from various levels of biological complexity (i.e. computational, in-vitro, and in-vivo).

This Research Specialist will be responsible for maintaining mouse colonies, genotyping, performing basic molecular biology and protein analysis including western blotting, protein expression, protein purification, etc. In addition, there will also be some lab management responsibilities that will include close work with the PI. Experience working with mice and basic genotyping skills are highly recommended. Organizational skills and significant prior experience in a research lab are strongly preferred.

The ideal candidate is a highly motivated and ambitious individual with a great passion for science that wants to join a team-oriented lab in an academic environment. This is a great opportunity for an individual interested in a long-term career in research. A minimum commitment of two years at least is desired. As the candidate becomes comfortable with daily laboratory tasks and maintenance, there will be opportunities to design and implement their own project and assist in data collection for grants and publications.

Duties & Responsibilities
  • Perform administrative functions such as filing, ordering of supplies, maintaining inventory, maintaining databases (for example: mouse colony, chemicals, MSDS, training records, and interface with RLSS).
  • Coordinate training of new laboratory members, including ensuring they have performed appropriate online and in person training as well as any appropriate lab tests.
  • Operate laboratory equipment and performs minor equipment maintenance. Communicates with service technicians for major equipment repairs or service.
  • Prepare buffers and reagents.
  • Cannulation and ex vivo “hanging” of mouse hearts for adult myocyte isolation (good hand-eye coordination required).
  • Perform basic molecular biology, western blotting, and protein purification.
  • Train students and staff in the operation of equipment and/or in the performance of technical procedures.
  • Analyze data and draft research progress reports including summarizing test results of assigned project.
  • Assist with data collection and generation of figures for grants.
  • Additional duties may be assigned as it relates to the position.

Bachelor’s degree in a field appropriate to the area of assignment AND two years related research experience; OR, Six years research experience appropriate to the area of assignment; OR, Any equivalent combination of experience, training and/or education.

Preferred Qualifications
  • Demonstrated ability to effectively communicate and work well with others.
  • Prior experience with handling and maintaining mouse breeding colonies, including transgenic mice and inducible mouse models.
  • Motivation to expand skill set and take on responsibility for lab management.
  • Willingness to acquire new skills, develop new techniques/protocols, and work independently.
  • Knowledge of principles and techniques of molecular and cell biology.
  • Strong organizational skills.
  • Word processing and data presentation skills (e.g. using programs such as Word; Excel; Prism).
